Creation of genetic constructs:

Bioinformatics development of genetic constructs:

 

This service includes designing primers that clone chosen DNA fragment, together with implementation of restriction sites, and simulating individual reactions of restriction and ligation during creation of genetic construct.

 

Physical creation of genetic cloning constructs:

 

This service includes development of the primers and their chemical synthesis, amplification reactions of the demanded DNA fragment, all required stages of DNA cleaning, as well as the implementation into the cloning vector. Result will be produced as ready-made genetic constructs in commercially-available plasmide vectors.

 

Physical creation of bacterial genetic constructs:

 

This service includes bioinformatics development of the primers and their chemical synthesis (on the basis of delivered sequences of the demanded insert and on the basis of the delivered biological material), amplification reactions of the demanded DNA fragment, all required stages of DNA cleaning, as well as the implementation into the cloning vector, selection of the expressive vector and cloning [from one to another].

 

Result will be produced as ready-made genetic constructs in commercially-available plasmide vectors that are implemented (on demand) into cloning or expressive bacteria delivered by the customer.

 

Remarks:

 

Material that is delivered by the customer:

  1. Matrix DNA, cDNA, RNA or a cloning construct
  2. Information on a demanded model sequence (CDS, miRNA, siRNA)
  3. Expressive bacteria (in case the prepared construct was to be implemented in the expressive bacteria)
  4. Expressive vectors (in case of implementation into a rarely-used vector)

 

Estimated realisation time depends on a difficulty level of constructs’ creation (amplicon’s softening temperature, primers’ binding sequence, length of DNA fragments that are to be cloned [into]) – it is 3 months, on average; together with cloning to the expressive vectors. This offer does not include optimisation of protein’s expression and changes in coding sequence, e.g. by a mutagenesis aiming to increase expression’s efficiency; [it includes] only the selection of expressive vector and expressive bacteria in order to optimise translation and correct protein’s folding.
